In 12th-century Japan, swordsman Kurou escapes ghastly pursuers into a deep forest with his servant Benkei. Superhumans who came into existence through different circumstances, and who are special in different ways -- Titans from outer space, lifeforms from a mystical world, phantoms and goblins from ancient times, cyborgs created by scientists, relics that rose out of the ruins of ancient civilizations, and the list goes on. One segment of these superhumans makes no secret of their existence and has gained popularity in society. Others cloak their identities and fight in secrecy.
There are enemies of superhumans as well, organizations that operate in the shadows.
The Japanese government has taken many measures to secure peace and order.
One is the establishment of an organization under the Ministry of Health and Welfare -- the Overpopulation Research Laboratory, a.k.a. the Superhuman Bureau.
Their mission is to identify and approach superhumans, then oversee and safeguard them. One member of the Superhuman Bureau, Jiro Hitoyoshi, is protagonist of this series.

The special will be a completely new work, featuring reanimated scenes from the beginning of the series through when the crew enters the Grand Line.
The special is commemorating the manga's 20th anniversary, and will also feature anime versions of the original manga's chapter cover stories as part of the special's ending credits. The staff haven't yet clarified which cover story or stories will be animated. Additionally, the anime will feature a special 20th anniversary commemoration version of the original opening theme song "We Are!".

Three young girls, Hikaru, Umi, and Fuu, are transported to a magical world called Cephiro during a field trip to Tokyo Tower. They are soon greeted by Master Mage Clef, who explains to them that they have been summoned to become the Legendary Magic Knights and save Cephiro. The girls are less than enthusiastic about this idea, and only want to return home.
Clef further explains that they must seek out the three Rune Gods to help them fight. He bestows armor and magical powers to each of them. They learn from Clef that High Priest Zagato has kidnapped the Pillar of Cephiro, Princess Emeraude. The Pillar of Cephiro has the sole responsibility of keeping Cephiro alive and in balance with her prayers. Without Princess Emeraude, Cephiro would fall into ruin. Hikaru, Umi, and Fuu must fight off Zagato's henchman and find the Rune Gods if they ever want to get back home. They soon learn that friendship and loyalty are the only things they can rely on in the crumbling Cephiro.

Ringleader of a bank robbery, Mi Liu is arrested and transferred to a special prison of the National Defense Agency that monitors vampires. Trying to break free with Anji, Mi Liu is attacked by strange monsters and he finds out that the prison location corresponds to the birthplace of vampires, the old capital Blue Town. Why are vampires trapped in Blue Town? What are those strange monsters attacking them? Our heroes must fight to solve those mysteries.

The story of the Hand Island no Bouken special is set a little before the events of One Piece Film Z and depicts a major "Hand Island" incident during the New World storyline.
